在pH 6.35 的 Tris-HCl 缓冲溶液中,头孢西丁与甲基绿反应生成具有两个明显正吸收峰的离子缔合物,最大和次大吸收波长分别位于664 nm 和560 nm,表观摩尔吸光系数分别为 2.98×104 和1.76×104 L/(mol.cm),头孢西丁的质量浓度在0.85~11.12 mg/L 以内遵守比尔定律。方法用于人体血液及药物中头孢西丁含量的测定,加标回收率在 99.1%~101.2% 之间,相对标准偏差在 1.6%~2.0% 之间,测定值与标示量一致。 |

In Tris-HCl buffer solution of pH 6.35, cefoxitin (CEXI) reacts with methyl green (MEG) to form an ion-association complex with two obvious positive absorption peak. The maximum positive absorption wavelength and sub-major positive absorption wavelength were 664 nm and 560 nm respectively, and apparent molar absorptivity (ε) were 2.98×104 and 1.76×104 L/(mol.cm), and Beer’s law was obeyed in the range within 0.85~11.12 mg/L of cefoxitin. The method was applied to determine the content of cefoxitin in commercially available cefoxitin sodium medicament and in human blood, and results obtained were in conformity with labeled values. Values of recovery by standard addition method was in the range of 99.1%~101.2% and RSD's (n=6) in the range of 1.6%~2.0%. |
