      通过化学还原法合成了球形银纳米颗粒,并利用电子显微镜方法对其形貌进行了表征;利用紫外可见吸收光谱法(UV)、荧光光谱法(FL) 以及扫描电子显微镜(SEM) 研究了球形纳米银与人血清白蛋白(HSA) 的结合反应。随着纳米银溶液浓度的增加,混合溶液的紫外吸收峰强度增加,但荧光强度则发生了明显的猝灭。光谱学实验结果表明,球形纳米银与人血清白蛋白在溶液中发生了相互作用,此结果也通过扫描电子显微镜实验得到了验证。由荧光实验还可获得纳米银与HSA 相互作用的结合常数、结合位点数以及吉布斯自由能变,由这些热力学数据可知纳米银与人血清白蛋白可以自发结合发生反应,并形成缔合物。

      A kind of Ag nanoparticles were successfully synthesized by the typical chemical reduction method, and the morphology of these nanoparticles was observed by using scanning electron microscopy (SEM) and transmission electron microscopy (TEM). Interaction of the Ag nanoparticles with human serum albumin (HSA) in aqueous solution has been investigated by UV-vis absorption spectroscopy, fluorescence spectroscopy (FL) and SEM. The absorbance of the mixture solution is progressively increased, but the fluorescence intensity is decreased with increasing concentration of silver nanoparticles. The spectroscopy results show that the interaction between the Ag nanoparticles and HSA occurs. Furthermore, the morphology change of the nanoparticles in the mixed solution was characterized by SEM experiments, which was in close agreement with that obtained by UV-vis and FL experiments. The association equilibrium constant (K), the number of binding sites (n) and the standard free energy change (ΔG°) were calculated by using the Stern-Volmer method. These thermodynamic data indicate that the binding processes of Ag nanoparticles and HSA are spontaneous, and the association complex can be formed in the solution.
