为合成得到高纯度高收率的5-甲基四氢叶酸,本文对传统的化学合成方法进行改进。即在反应液中加入保险粉,且以柠檬酸-磷酸氢二钠缓冲溶液替代单纯的水溶液。并利用高效液相色谱检测手段检测反应,确定硼氢化钠的添加量、反应温度、缓冲液浓度和pH值、甲醛添加量等因素对反应的影响。结果表明:硼氢化钠的总添加量为0.65g、反应温度为50℃、缓冲液浓度为0.2M和pH值为6.5、甲醛添加量为0.4mL时,目标产物纯度达88%,收率达70%。 |

In this paper, we have established a novel method to synthesize 5-methyl tetrahydrofolate (5-MTHF) with high purity and high yield, by introducing sodium hyposulfite into the reaction and also substituting citric acid-disodium hydrogen phosphate buffer solution for water. We carried out high performance liquid chromatography (HPLC) to check the purity and researched the influential factors of the reaction that including the addition of sodium salt, reaction temperature, buffer concentrat-
ion, pH values, etc. The results revealed that the purity of the target product was almost 88.9% and the yield was 70.4%, which could be realized when the folic acid amount was 0.2g, NaBH4 amount was 0.65g, the reaction temperature was 50℃, the concentration of the buffer was 0.2M, pH value was 6.5 and the addition of formaldehyde was 0.4mL. |
