本文利用巯基化合物对异硫氰酸荧光素(FITC)的荧光淬灭作用,建立了一种快速检测巯基化合物的新方法。我们在实验中发现,巯基化合物能够淬灭FITC的荧光,根据此现象,发展了检测巯基化合物的新方法。本方法的用时非常短,在优化了pH、反应时间等条件后能在5分钟之内完成对巯基化合物的检测。在最佳条件下对实际样品的测试取得了很好的效果,且具有良好的选择性。对谷胱甘肽(GSH),高半胱氨酸(Hcy),半胱氨酸(Cys),二硫苏糖醇(DTT)的检测限分别达到3.08μM ,2.66μM,1.28μM ,1.92μM。 |

In this work, a novel method for rapidly detecting thiol compounds was established using fluorescein isothiocyanate (FITC) as fluorescence probe. The method was based on the quenching of the fluorescence of FITC by thiol compounds. Thiol compounds could be detected in 5 minutes at the optimized conditions including pH and reaction time. The method was used to detect glutathione(GSH) in real samples and showed good selectivity and recovery. The limits of detection (LOD) were 3.08μM for glutathione (GSH), 2.66μM for homocysteine (Hcy), 1.28μM for cysteine (Cys), and 1.92μM for dithiothreitol (DTT). |
