Vidarabine (Ara-A) is a broad-spectrum DNA virus inhibitor, which has been used clinically for the treatment of herpes simplex encephalitis, and also be used for treat herpes zoster and varicella from immunosuppressed patients infected. Its derivatives such as Vidarabine Monophosphate and Fludarabine are also effective in the treatment of related diseases. The preparation method comprises: chemical synthesis and biosynthesis. in the early 1960s, Vidarabine was first successful synthesized through chemical synthesis. Subesequently, In the late 1960s, Vidarabine was obtained with enzymatic method in biosynthesis. The characteristic of chemical synthesis is a wide range of raw materials, and synthesis method varies; biosynthesis research focused on enzymatic methods, and the key process is the preparation of enzymes. Since both the two approaches have their advantages and disadvantages, it can be used in the practical production. The latest progress in the preparation of Vidarabine was reviewed and the prospect was also discussed in this paper. |
