      建立了基于铜纳米簇(copper nanoclusters,CuNCs)的免标记荧光生物传感器快速检测链霉素的方法。利用链霉素核酸适配体和双链茎设计并合成了发夹结构DNA(hairpin DNA,hpDNA)模板,富含AT的双链茎部分可以合成CuNCs。在CuNCs荧光传感体系中加入链霉素后,链霉素与核酸适配体结合,使hpDNA双链茎打开,CuNCs荧光强度明显下降。在最优条件下,该方法检测链霉素的线性范围为0.1~20 mg/L,线性回归方程为y=240.17-10.31x,线性回归系数r2为0.9954,检出限为4.36 μg/L。该方法对牛奶样品的加标回收率为98.6%~104.8%。方法无需复杂的DNA序列设计、荧光标记,实验操作简单易行,适用于链霉素的快速检测。
英文摘要:
      A rapid and label-free fluorescent sensing detection method has been developed for streptomycin by using copper nanoclusters biosensor. The hairpin DNA (hpDNA) template contained a streptomycin aptamer loop and a AT-rich double strand stem, and the AT-rich sequences are the template for the formation of fluorescent CuNCs. When streptomycin is present in the hpDNA-CuNCs solution, the fluorescence intensity decreased effectively, due to the structure switching of the hpDNA. Under optimized conditions, the method showed a good linear relationship with the streptomycin concentration in the range of 0.1-20 mg/L with correlation coefficients (r2) 0.9954. The detection limit of this method was 4.36 μg/L. Furthermore, the method was successfully applied to detect the streptomycin in milk samples and showed recoveries ranging 98.6%-104.8%. The method didn’t need complex DNA sequence design and was free of fluorescent labeling, and the operation is simple. Therefore, it is suitable for the rapid detection of streptomycin.
